After an experiment, you might want to store the probe for further uses.
There are two possible ways to store your used FluidFM probes depending on how they were used:
If used in liquid: store in liquid.
If used in air: store in humid air.
Appropriate storage containers
We recommend the following containers for our FluidFM probes, depending of the platform and probe form-factor:
FluidFM ADD-ON for FlexAFM and Nanowizard: 24 well plate.
FluidFM ADD-ON for BioScope Resolve: Blister used for shipping of probes.
FluidFM BOT: 12 well plate with fitting storage adapter (accessory available form Cytosurge).
Store in liquid:
The probe was used in a liquid medium such as cell buffer or water. It now should also be stored in water so that drying out of the probe will not deposit crystals or contaminants on the probe’s surface, eventually clogging the aperture.
Store the probe in distilled, filtered water with added 2% antibiotic and antimycotic solution (Sigma-Aldrich, A5955).
Put a lint-free tissue on top of the stored probes, then wet it with lab-grade water. Close the lid of the 12 or 24 well plate on top of the wet tissue.
Make sure the probe never dries out during the storage period and refill the reservoir of the probe (with lab-grade water) periodically, this ensures that the probe stays hydrated, while the reservoir is not diluted.
The antibiotic and antimycotic will prevent the formation of biofilms that would eventually clog the FluidFM probe. Storing in the fridge at 4°C is recommended.
Storage of up to 1 week can be achieved with this protocol.
Please note that water soluble coatings (if any) have to be reapplied for the next experiment.
Store in humid air:
For spotting in air, the probe is typically filled with glycerol or another liquid which does not evaporate quickly. In this case, the probe can be stored in a highly humid environment: place the probe back in the original blister and enclose it in a hermetic container together with a humidified Kimwipe. Also here storage in 4°C is recommended for minimal evaporation.
